New monoclonal antibody developed that can target proteins inside cancer cells

Mar. 13, 2013 ? Researchers have discovered a unique monoclonal antibody that can effectively reach inside a cancer cell, a key goal for these important anticancer agents, since most proteins that cause cancer or are associated with cancer are buried inside cancer cells. Scientists from Memorial Sloan-Kettering Cancer Center and Eureka Therapeutics have collaborated to create the new human monoclonal antibody, which targets a protein associated with many types of cancer and is of great interest to cancer researchers.

Unlike other human therapeutic monoclonal antibodies, which can target only proteins that remain on the outside of cancer cells, the new monoclonal antibody, called ESK1, targets a protein that resides on the inside of the cell.

ESK1 is directed at a protein called WT1, which is overexpressed in a range of leukemias and other cancers including myeloma and breast, ovarian, and colorectal cancers. WT1 is a high priority target for cancer drugs because it is an oncogenic protein, meaning that it supports the formation of cancer. In addition, it is found in few healthy cells, so there are less likely to be side effects from drugs that target it.

"This is a new approach for attacking WT1, an important cancer target, with an antibody therapy. This is something that was previously not possible," said David A. Scheinberg, MD, PhD, Chair of the Sloan-Kettering Institute's Molecular Pharmacology and Chemistry Program and an inventor of the antibody. "There has not been a way to make small molecule drugs that can inhibit WT1 function. Our research shows that you can use a monoclonal antibody to recognize a cancer-associated protein inside a cell, and it will destroy the cell."

The first studies of the antibody are showing promise in preclinical research as a treatment for leukemia as reported March 13, 2013, in Science Translational Medicine.

"ESK1 represents a paradigm change for the field of human monoclonal antibody therapeutics," said Cheng Liu, PhD, President and Chief Executive Officer of Eureka Therapeutics. "This research suggests that human antibody therapy is no longer limited to targeting proteins present outside cancer cells, but can now target proteins within the cancer cell itself."

ESK1 was engineered to mimic the functions of a T cell receptor, a key component of the immune system. T cells have a receptor system that is designed to recognize proteins that are inside the cell. As proteins inside the cell get broken down as part of regular cellular processes, molecules known as HLA molecules carry fragments of those proteins -- known as peptides -- to the surface. When T cells recognize certain peptides as abnormal, the T cell kills the diseased cell.

In the current study, the investigators showed that ESK1 alone was able to recognize WT1 peptides and kill cancer cells in the test tube and also in mouse models for two different types of human leukemia. "We were surprised that the antibody worked so well on its own," said Dr. Scheinberg, senior author of the paper. "We had originally expected that we might need to use the antibody as a carrier to deliver a drug or a radioactive therapy to kill the cancer cells, but this was not necessary."

Additional studies must be done in the laboratory before ESK1 is ready to be tested in patients. But the monoclonal antibody was engineered to be fully human, which should speed the time it takes to move the drug into the clinic. Researchers expect that the first clinical trials, for leukemia, could begin in about a year.

The antibody was developed under a collaborative effort between Memorial Sloan-Kettering and Eureka, which have jointly filed for patent protection.

This work was supported by grants from the Leukemia and Lymphoma Society, the National Cancer Institute, the Sloan-Kettering Institute's Experimental Therapeutics Center and Technology Development Fund, the Commonwealth Foundation for Cancer Research, the Tudor and Glades Foundations, the Merker Fund, the Lymphoma Foundation, and the Mesothelioma Applied Research Foundation.

New BlackBerry coming to the US public on March 22

(AP) ? BlackBerry-maker Research In Motion will begin selling its new touchscreen smartphone to U.S. consumers with AT&T on March 22. The release will come several weeks after RIM launched the much-delayed devices elsewhere.

AT&T said Monday that the Z10 will be available for $199.99 with a two-year contract. Sales of the device began in the U.K. and Canada shortly after RIM unveiled the phone in late January.

Rival U.S. carrier T-Mobile said it expects to deliver the new BlackBerry for some corporate customers as soon as the end of this week, though it did not provide details on the availability for non-business customers.

The redesigned BlackBerry is RIM's attempt at a comeback. The pioneering brand lost its cachet not long after Apple's 2007 release of the iPhone, which reset consumers' expectations for what a smartphone should do.

RIM Chief Executive Thorsten Heins said previously he was disappointed the new BlackBerry would not be released in the U.S. until mid-March, but he said the U.S. and its phone carriers have a rigid testing system.

Heins told The Associated Press last month that the company would have to regain market share in the U.S. for BlackBerry to be successful. The U.S. has been one market in which RIM has been particularly hurt. The iPhone and phones running Google's Android software now dominate. According to research firm IDC, shipments of BlackBerry phones plummeted from 46 percent of the U.S. market in 2008 to 2 percent in 2012.

Heins also suggested to the AP that a modern BlackBerry with a physical keyboard might not arrive in the U.S. until May or June, a month or two behind other parts of the world. Heins said the physical keyboard version, the BlackBerry Q10, will likely come out eight to 10 weeks after a carrier releases a model with only a touch screen, the BlackBerry Z10. With the Z10 set for release in the U.S. on March 22, eight to 10 weeks brings the U.S. date for the Q10 to mid-May to early June.

Shares jumped $1.84, or 14.1 percent, to close at $14.90 Monday on the Nasdaq. BGC Financial analyst Colin Gillis said the stock move is based on a report quoting Lenovo's chief executive as saying he might be interested in an acquisition of RIM. Gillis and Jefferies analyst Peter Misek said a Chinese acquisition is unlikely due to security concerns.

Misek said checks by Jefferies indicate that the Z10 is selling well in developing markets including India, but Gillis noted RIM's U.S. release will go up against Samsung's next Galaxy smartphone which is expected to be unveiled on Thursday.

"If that makes the splash that people think it may, you don't want to be the guy that's coming out a week later," Gillis said.

Publishers represented by the Authors Guild and Association of American Publishers have filed objections to Amazon's pursuit of new generic top-level domains ".book," ".author" and ".read." While some of those gTLDs have already come under fire from entire countries, the influential book groups told gatekeeper ICANN that "placing such generic domains in private hands is plainly anticompetitive," adding that it would allow "already dominant, well-capitalized companies" to abuse their market power. ICANN plans to assign rights to organizations or companies to manage domain suffixes like the current ".com" or ".org" and firms like Google, Microsoft and Amazon have sought names like ".app" and "movie," often in competition with each other. Competitor Barnes & Noble filed its own protest, saying that Amazon "would use control of these TLDs to stifle competition in the bookselling and publishing industries." If such protests are persuasive enough, companies could lose not only the domain name in question, but 20 percent of the $185,000 application fee -- admittedly pocket change for outfits like Amazon.

Russian police arrest 3 suspects in Bolshoi attack

MOSCOW (AP) ? Russian police said Tuesday they arrested three men in the acid attack that nearly blinded the artistic director of the Bolshoi ballet, including a star dancer suspected of masterminding the plot.

Sergei Filin was left with severe burns to his eyes and face when a masked attacker threw a jar of sulfuric acid in his face as he returned home late on Jan. 17. The 42-year-old former dancer is now undergoing treatment in Germany.

Interior Ministry spokesman Anatoly Lastovetsky said Bolshoi soloist Pavel Dmitrichenko is suspected of planning the attack.

Police also arrested the suspected perpetrator of the attack and the man believed to have driven him to and from the scene, the spokesman said.

It was unclear whether either man had any connection to the famed ballet company.

Dmitrichenko, who joined the Bolshoi in 2002, has danced several major parts in recent years, including Ivan the Terrible in the ballet of the same name.

Bolshoi Theater spokeswoman Katerina Novikova said management was not aware of a conflict between him and Filin. Channel One state television reported that Dmitrichenko's girlfriend, however, also a Bolshoi soloist, was known to have been at odds with Filin.

The Bolshoi Theater is one of Russia's premier cultural institutions, best known for "Swan Lake" and the other grand classical ballets that grace its stage. But backstage, the ballet company has been troubled by deep intrigue and infighting that have led to the departure of several artistic directors over the past few years.

Filin's colleagues have said the attack could be in retaliation for his selection of certain dancers over others for prized roles. Filin told state television before he checked out of a Moscow hospital that he knew who ordered the attack but would not name the person.

The Interior Ministry said the alleged perpetrator, 35-year-old Yury Zarutsky, was arrested in the Tver region north of Moscow. Earlier in the day, police had detained and questioned the suspected accomplice, whom the spokesman identified as Andrei Lipatov. No other details were provided.

Russian news agencies reported that Lipatov had been detained in the town of Stupino, a sprawling Moscow suburb that has summer houses owned by the Bolshoi Theater and used by its dancers and management. Dmitrichenko said in a recent interview that he was managing the dachas in his spare time.

In a February interview with Snob magazine, the Bolshoi's general director, Anatoly Iksanov, said the attack on Filin was inspired by longtime principal dancer Nikolai Tsiskaridze.

"I don't blame that particular crime on him, but I'm accusing Nikolai of escalating the situation at the theater, of putting psychological pressure on the theater's staff and management, on Filin, on myself and teachers," he said.

Tsiskaridze, a long-time critic of the theater's management, has denied the allegation and accused Iksanov and his allies of fueling the dispute.

Dmitrichenko's girlfriend, Anzhelina Vorontsova, was coached by Tsiskaridze.

Many ballet stars, including Anastasia Volochkova, have sided with Tsiskaridze. Alexei Ratmansky, the Bolshoi ballet's artistic director from 2004 until 2008, has likened the atmosphere at the theater to "a disgusting cesspool" and said that the attack stems from "the lack of any ethics at the theater."

Ratmansky is now an artist-in-residence at the American Ballet Theater in New York.
